Partai yang didirikan di Ripon Wisconsin pada 28 Februari 1854, sebagai sebuah partai yang melawan perbudakan dalam wilayah baru. Partai ini tidak boleh disamakan dengan Partai Demokratik-Republik AS-nya Thomas Jefferson atau Partai Republik Nasional AS-nya Henry Clay.
Konvensi pertama Partai Republik diadakan pada 6 Juli 1854 di Jackson, Michigan. Banyak dari kebijakan awalnya terinspirasi dari Partai Whig AS. Banyak dari anggota awalnya berasal dari Partai Tanah Bebas (Free Soil Party) dan Partai Amerika. Sejak didirikan, oposisi utamanya adalah Partai Demokrat.
Simbol resmi Partai Republik adalah gajah. Meski gajah telah sering dikaitkan dengan partai tersebut, sebuah kartun politik karya Thomas Nast, yang diterbitkan di Harper's Weekly pada 7 November 1874, dianggap penggunaan penting pertama simbol tersebut. Pada awal abad ke-20, simbol tradisional Partai Republik di negara-negara bagian barat tengah seperti Indiana dan Ohio adalah elang, berlawanan dengan ayam jantan yang digunakan Demokrat. Simbol ini masih tampil dalam kertas suara di Indiana.
